Ipamorelin is a synthetic growth-hormone secretagogue that activates the ghrelin / GHS-R1a receptor. Instead of supplying growth hormone directly, its appeal is that it stimulates the pituitary to release a pulse of the body’s own GH.

The “Pulse” Is The Whole Point

Human volunteer studies confirmed that ipamorelin can produce a discrete episode of GH release. That pulse-like behavior is central to its popularity: users are generally trying to increase GH signaling while retaining some of the body’s normal feedback and pulsatility.

In a human pharmacokinetic/pharmacodynamic study, GH peaked roughly 0.67 hours after administration and then declined, while ipamorelin showed a short terminal half-life of about two hours.

Ghrelin Receptor → Pituitary GH Release

Ipamorelin binds the growth-hormone secretagogue receptor and stimulates somatotrophs in the pituitary. The downstream interest is familiar GH-axis biology: GH can increase hepatic and peripheral IGF-1 signaling and influence protein metabolism, lipolysis, tissue repair and growth.

We Know It Can Raise GH; The Rest Is Less Certain

Controlled human pharmacology demonstrates GH release, but that is not the same as proving better sleep, improved recovery, muscle gain, fat loss or anti-aging outcomes in otherwise healthy adults. Those real-world goals have a much thinner direct evidence base.

Ipamorelin is not FDA approved. FDA has specifically noted limited safety information for compounded injectable ipamorelin and concerns related to peptide impurities, aggregation and immunogenicity. Route also matters: safety findings from one route cannot simply be assumed to apply to another.
